| | A. | In terms of first choice I would have suggested the folmeterol first, it has much fewer side effects than theophyllines. Though once upon a time I was a fan, ( many years ago!) theophyllines have a very narrow therapeutic window and, even in patients who tolerate them for a while often there are late side effects such as nausea, muscle cramps and abdominal pain. Ideally also blood levels should be checked periodically to ensure that they do not become toxic.
Folmeterol has a large range of dose related response, for the 12 microgram unit the daily dose for regular use should not exceed 4 inhalations, however occasionally up to a maximum of 6 inhalations may be allowed within a 24-hour period. | | | |

| | A. | Theophylline is 1,3-dimethyl-7H-purine- 2,6-dione and Caffeine is 1,3,7-trimethyl-1H-purine-2,6 (3H,7H)-dione. Look carefully at these structures and look at the difference in what is being dissociated for the Kd values.
As for the HPLC, you did not specify if you were using a normal phase or reverse phase (C-18) column. Was the mobile phase only water, methanol and/or acetonitrile or does it contain acetic acid?
In a typical C-18 reverse phase HPLC analysis, the non-polar compounds adsorb into the column longer than the more polar compounds (which remain in the mobile phase longer and so elute first). | | | |
